We're going to the keys in March too, its our first time there, so I don't know much about answering your questions. What I will advise though is to book accommodation early, as it is spring break and all the best resorts get booked solid. We have had ours booked since last summer - nothing like forward planning :D

It takes about an hour and a half to get form Miami to Key Largo, and about 3-4 hours depending on drive speed, stop offs to get from Key largo to Key West...where are you thinking of staying on the Keys.

Depends on where you prefer to stay, large resort style hotels or smaller old town gingerbread b & bs ?
My favourite is www.edenhouse.com located within walking distance of the main drag of Key West bars and the harbour area. There are plenty of similar places such as CoCo Plum Inn, Almond Tree House, Duval House, Nassau House and dozens of others. Be aware that some of the others you will find popular on www.tripadvisor.com may cater mostly to gay visitors, not saying that's a problem but it's worth knowing.
If booking smaller hotels it can be best to deal with them directly by phone or e-mail rather than through travel websites such as Expedia or Hotels.com. For the larger chains sometimes the reverse is true as deals can vary.
If travelling down from Orlando a stopover in Miami breaks up the journey nicely, you can stay on South Beach itself at the Holiday Inn for around $80 including free parking, within walking distance of Lincoln Road Mall and the beach front bars / clubs.
I would try to stay three nights in Key West rather than two.

We have booked to stay at www.pinesandpalms.com on Islamorada. This is about an hour and a half south of Miami. From here we intend to visit www.johnpennekamp.com an underwater state park, perfect if you enjoy snorkelling.
I can recommend the following website for information and accommodation on the keys. We found a lot of useful info on here.

www.fla-keys.com

May see you down there? :p:

We stayed in Islamorada after 9 days in Disney, enjoyed it purely as a wind-down. There is a lot of driving to get to Key West and its all road, not particularly scenic (unless you like just gazing at the sea!). It's a fishing and diving paradise though, so if you do these things fine, if not you may find it a bit boring (after being over stimulated at Disney, we found sitting around all day was a drag!) Also, we thought that cafes/restaurants were a bit limited and all had the same menu (fish, chicken, steak, fish). Key Largo is an absolute dump and to be avoided. It's at the beginning of the keys and you have to drive through it to get anywhere else so you'll see what I mean then!
