The obvious route is train and tube but I guess you don't want to do that with kids and luggage - especially the tube part. You could get the tube to Paddington and then the Heathrow Express but again, that could be too stressful with kids.
One alternative could be to use the Cross Country trains from Derby to Reading and then to get the RailAir coach into Heathrow. It's not the fastest but you can get combined tickets which save money I think.

We got the train too and from Gatwick with 4 huge cases, 4 backpacks, 2 kids and a buggy! We went up the night before and did the twilight check-in and stayed at the premier inn.
Coming home was harder because we were shattered from not sleeping on the overnight flight home. Personally I would say get a train there and a taxi home again, getting a friends and family railcard will reduce the train cost greatly |

We are in Nottingham and did train to St Pancras then tube to Heathrow.
It was fine, the tube takes about an hour but was super cheap as we travelled off peak on the way down. Biggest hassle was on the way back, hanging about at St Pancras as we were booked on a specific train and chose a later one as the price was loads cheaper. This was in November so it was cold but not too bad if it’s warm. 😁
