I live in Singapore. Have been to KL before but haven't been there for a long time so won't attempt to give any information on KL! Will however add that in general the cost of things in Singapore is higher than in Malaysia.
Singapore is very well connected to most of the rest of South East Asia, and there are plenty of flights going to most nearby destinations. Some nearby popular destinations (where people I know have been on weekend trips) include Bali, Bangkok, Chiang Mai, Penang, and a bit further afield would be places like Hong Kong and Taiwan (both about 4-5 hours away). I know in the past that flights with Singapore Airlines for an itinerary like London-Singapore-Penang-Singapore-London (with stopover in Singapore) wouldn't have cost much more than just London-Singapore-London, but I'm not sure if this is still the case. Happy to answer questions on Singapore if you have any!

When we went to Singapore we paired it with Thailand, Khao Lak and Phuket. We got really cheap flights with Silk Air between Singapore and Phuket, (flew from the uk with Singapore Airlines) that might be an option if you were interested in Thailand.

It was many years ago when we visited. We stayed in KL at the Shangri-La hotel for 3 nights and found that enough time, great hotel with rooftop pool. Kuantan we stayed for 5 days and then 5 days at Pankgor Laut Island, which was just heaven on earth. I agree with others that maybe you would be better sticking to two locations with the days you have, although Im sure it is doable if planned well.
