We had a fabulous time in Toronto and would go back tomorrow.
So that people can advise you better, what time of year are you thinking of going and how long for?

We stayed in Toronto for 2 nights as part of our honeymoon, and as well as a trip to Niagra from there, we went up the CN Tower, which is one of the major landmarks.

Is a fab place. Would recommend hop on/off bus tour to get your bearings. Distillery district is nice, Eaton centre fab shopping centre, several chinatowns, Kensington Market and my favourite is a boat trip to Toronto Islands (sadly currently suffering flooding but think that is v unusual. Loads of food choices, my favourite is a place called Bannock. Try yo make sure you get to see Niagara on the Lake - small town used for some of filming of Its a Wonderful Life.

It is a lively city - lots to do. We stayed a week there and were never bored!
You could combine it with a week in a cottage by the water in Ontario or even do a circuit down to NYC. There are quite a few trip reports on Toronto which would be worth a look for ideas!

We went at the end of May, so a similar time of year.
We enjoyed the CN Tower, Toronto Islands, the Distillery District, Casa Loma, Niagara Falls, a baseball game and a brewery tour amongst other things. There's a link to my trippie in my signature if you're interested. Quite a few of us went at a similar time, so if you do a search, you'll find other trippies too. I found them invaluable in helpful me to plan my trip. As others have mentioned, Niagara on the Lake seems to be worth a trip, bit we only had time for a quick drive through it. If I went again, I'd do a split stay and stay for a night or two in the Niagara area to make the most of that.

As a few replies have already mentioned, the CN Tower (check the fixed price dinner menu at the 360 restaurant which also gives admission up to the viewing platform), Distillery District and Casa Loma.
We used the hop-on/off bus tour that gave a great overview of the city, and we also used it to visit Casa Loma (which is a distance from the centre). It also had the bonus of a boat trip out into the bay and around the islands. Another vote for Niagara on the Lake - this was an absolutely beautiful town, so far removed from the commercialism of downtown Niagara Falls. Worth renting a car to get there as it wouldn't be that easy using public transport, although some day tours (I think) call there. As someone else has mentioned, it could be worth visiting Niagara then staying over in Niagara on the Lake, maybe the night before you return to the UK (if you are only in Toronto for a short visit) as the airport is on that side of Toronto. When we stayed, we rented a condo through Airbnb which was located about 10 mins walk behind the CN Tower in the Entertainment District - great location and much more room than a hotel room (as well as being cheaper). We also did a trip report which may be of use to you - it's the "East and West is Best 2" report.

We stayed a total of 3 nights in Toronto last year. We've got another night there this year (was hoping for 2 nights but couldn't squeeze it in).
Not much to add to what everyone else has said, but for us the must-do's were the CN Tower, a night in Niagara Falls, the hop on/off bus tour (which also gives you a free boat trip around the islands.). Our DS loved the Ripley's Aquarium which is next to the CN Tower. I loved just walking the streets - it's a lovely, clean city. On the way out of Niagara we also stopped in Niagara On The Lake, which was absolutely lovely but we didn't get to spend enough time there. This year we're hoping to get to Casa Loma and one of the museums in our not-quite-2 days there.
