We'll be flying to Sanford the year and are staying in the Davenport area.

Anyone know how long it will take in the car?

The route papamouse has suggested is the most direct route, however it's straight along I4, which can become a bit of a parking lot early evening. Depending on what time you arrive, you might want to consider using the 417 toll road, from just outside the airport to I4 past Disney.

We'll be using a friend's SATNAV (such a godsend last time!)

We've got a AoA Thomson villa, hope we can find where to get the keys from a bit more easily than last time
