To be honest I think a day is pushing it from Orlando. Try to get an overnight stay and make the most of it. Amazing place and so much to see and do - we had a couple of nights and left loads to see in the future.
our trip report days can be read here and here if it is any help to you - even if you are there for a day I'm sure you will be back there again one day Hidden Floridian gem

We have actually done it as a day trip but there was only the two of us and I guess we only explored the centre. It's about 2.5 hours from Highgate where we stay.
St Augustine is the oldest continuously settled European community in the USA. We parked at the Old Jail free and caught this trolley. trolleytours/st-augustine Mick

Maybe a long drive just for a day but if you are happy with travelling that distance it is a lovely place to visit. I would also recommend using the trolley tours as they take you to all the sights & you can hop off & on as much as you like & choose what you are interested in. Good to get rid of the car as soon as you enter the town. The Spanish part of the town is lovely pedestrianised area with lots of choice to eat & a nice walk along the waters edge. We stayed near st Augustine a few years ago so were able to have several trips into town to see everything using the trolley each time

It is certainly do-able in a day if you don't mind a long day. But, as Walt4it points out, an overnight stay will really give you the best of it. We also have a video highlights trip here:
