Sorry OP, I can't see where you're coming from. I've only ridden Single Rider as a single rider (i.e. I've only ever entered the line alone), and been regularly surrounded by groups and families in that line. It has never occured to me that this is an issue?
As others have said, Single Rider lines aren't really a way for guests riding alone to get to ride more quickly, it's a way to ensure ride vehicles leave full every time - which keeps overall wait times down.
I wouldn't care if a group of 100 was in the Single Rider line in front of me as long as when they get to the front they accept potentially riding in 100 different ride vehicles.
I think perhaps you've misunderstood the point of Single Rider lines?

The CM always tries to explain that this is single rider, but what can they do if the parent is adamant and won't budge without going together. Single riders should be separated in my opinion, but you can queue with as many family and friends as you like.

What can they do ? They can send them back to the standard line if they insist on riding together .

Can't see the problem myself, it should be an issue.

We love single rider at Universal, get to do so much more as we are not fussed about being next to each other there

As others have stated, the point is that you will probably not RIDE together. Nothing says you can't QUEUE together. Maybe you have misunderstood the purpose? Don't worry, be happy

thats exactly what we use single rider for too - we are a group of adults and older teens, but we are riding as singles. I've not once gone through the regular line for Men In Black for example, have always done single rider for that ride.

I rode Gringott's as a single rider last October many times, once or twice I had to wait a while to get on the ride. There were a few of occasions when couple's were in front of me were trying to get on together but they weren't allowed by the CM. On Spiderman, I remember myself, son and daughter all queuing together but not riding together - so, nothing wrong with families going into single rider lanes in my opinion.

this happened to me at Thorpe park, the single rider queue was as long as the main queue for the Saw ride...as far as I could see I was the only single rider and huge groups of people. I was really annoyed as being the only genuine single rider with huge groups of 6 and more clearly in the queue! very irritating but not something you could stop I don't think

I don't understand how it's irritating - the line is for people willing to ride as a single and fill in the spaces on the ride - what difference does it make if they know other people in the queue?
The only annoyance is those who try to insist on riding together.

why do groups insist on the single rider line? surely they only do it because they think they are going to ride quicker than the 'main' queue?

That's definitely why we (usually just 2 of us) do it and I'm guessing why most do.
